Hidden genetic timebomb for early bowel cancer discovered by scientists

Discovering why some people face a high bowel cancer risk young is vital. A groundbreaking study from Taiwan has unearthed new clues. Scientists created the region's first large-scale genetic database for colorectal cancer (CRC).

Unlocking hidden dangers

Hereditary colorectal cancer (HCRC) makes up 5-10% of all CRC cases. It significantly influences family risk and early diagnoses.

Experts found 24 pathogenic variants in HCRC-related genes. This was among 5.2% of Taiwanese CRC patients, with the figure rising to 8.2% in early-onset cases.

CRC isn't always down to chance. Genetic factors play a big part.

Up to 30% of patients report a family history. Also, 5-10% carry pathogenic variants passed down through generations.

These include conditions like Lynch syndrome and Familial Adenomatous Polyposis (FAP). Early screening for these variants is vital for prevention.

Why this matters for you

The study highlighted novel variants. One striking case involved a 26-year-old with a large 11 cm tumour. This patient carried a variant in the POLD1 gene.

The findings advance our understanding of genetic risk within the Taiwanese HCRC population.

Researchers believe many population-specific pathogenic variants are still hidden. They found 25.5% of rare variants were previously undocumented. The study underscores an urgent need to expand genomic databases for Asian-specific variants.

Yi-Chen Huang, the study’s first author, highlighted the broader implications. She stated: “Moving forward, we are working to characterize these candidate pathogenic variants by predicting changes in protein structure and integrating multi-omics data, with the goal of maximizing their clinical impact through bioinformatics.”

Co-corresponding author Han-Mo Chiu added his insights. He said: “This study identifies germline risk factors that enable earlier detection and precision care in CRC, while helping to close the gap in defining germline risk in Asian populations, particularly in early-onset CRC, which is rapidly increasing in Taiwan.”
